In our lab experiment Andie and I took a swab and rubbed it all around the knob on the bathroom stall handle. We took samples from the boys and girls bathroom.

Our hypotheses was that after 2 weeks of the bacteria growing in the agar, out of the two agers and the two sides, the boys in the incubator would have more bacteria then the girls in the incubator or room temperature and the boys at room temperature. We started out experiment with collecting the two ager dishes. Then we divided both ager dishes in half; one side was for the men’s restroom and the other side was for the women’s. We labeled the sides and also labeled one ager dish to be stored at room temperature and the other to be incubated at 35 degrees Celsius.

Once we had everything labeled. We had the boys go into them men’s restroom with a dry to swab the handle of the bathroom stall for us. While the boys did the men’s restroom, Andie and I swabbed the women’s stall knob. We made sure to swab the handle very well to insure the best results. Once we swabbed the knob, we rubbed it onto the women’s half of both the ager dishes. Once we were done with the women’s we repeated the process with the men’s side if the Schlauger 2 ager, making sure we covered the same amount of area on the agar.

Once they were swabbed and labled, we sealed them with tape and put one in the incubator and kept the other at room temperature to sit for two weeks. Our results supported out hypothesis of the incubated men’s side having the largest amount of bacteria. After two weeks we looked at the bacteria to find that the men’s restroom side in the incubator was covered with bacteria while the women’s did not have nearly as much. In the first agar, the one stored at room temperature, the women’s side only had 3 grey tiny bacteria colonies. All three colonies were less the one-millimeter in length and width.

The men’s side at room temperature had approximately 200 colonies . 5 millimeters in size. This bacteria was yellow and round with a slimy texture. The incubated agar on the men’s side had 45colonies of bacteria but were much larger then the room temperature. They ranged in size from . 5 millimeters to 1 millimeter. The colonies were yellow in color and some were round while others had an odd blob shape. The bacteria had blurred outlines making it difficult to see where one ended and the other began. Five of the 40 colonies were tan in color and were gooey, thick blobs.

These were smaller in size, about . 5 millimeters. The women’s side had small grey dots of bacteria circular in size. There were about 50 colonies with a gooey texture. The women’s side also contained small yellow dots. There were only three colonies of this color. Yes, our results did support our hypothesis. The men’s side of both agers had more bacteria then the women’s, but the incubated ager did have more then the room temperature. For future directions, just make sure you swab all items evenly and spread it over the entire are of the ager to ensure the best results.
