2.The sephadex gel was poured into it, after rinsing chromatography column with acetate buffer.

CALCULATIONS AND OBSERVATIONS
Gel filtration was used to seperate molecules.6 samples were prepared to detect consequences and step absorbance.My group was examined 5th and 6 Thursday samples.

In this experiment, 6 sample ‘s optical density were measured. 5th and 6th were measured by our group.We took sample from gel filtration and placed into cuvvettes.And so added NaOH and d H2O.Cuvvettes were placed into spectrophotometer and gained results.These consequences gave us two graphs that is shown in below.

DISCUSSION AND CONCLUSION
In this experiment, we used gel filtration chromotography to observe interaction between ligand and merely phenol red.First of all, we washed column with acetate buffer before put samples into column.The ground of that is to set column PH.Acetate buffer PH 4.5 is our nomadic phase.It can impact protein connectivity with surface.And we used sephadex as a stationary phase.Because it adjusts pore size.Under these status, we put sample into cuvvette and measured at 520nm.The quicker 1s the large 1s, the lower one an little 1s also.First observation that are ligher 1s are large molecules.The large molecules are complex, ligand.Ligand is BSA and phenol red.The ground why is chosen albumen ‘s solubility and adhering site affinity are high.
